The binding of allergens to IgE on mast cells triggers the release of several mediators, of which histamine is the most prevalent. Histamine is mainly produced in the tissue mast cells and blood basophils as a consequence of antigen challenge or tissue damage. More than 40% of all prescription drugs are thought to act against GPCR integral membrane protein targets2. The GPCR "Superfamily" with a collection of proteins with structural and functional characteristics in common, whose protein sequences share greater than 20% sequence identity in their transmembrane regions, and is presumed to have evolved from a common ancestor. While primitive organisms like yeast have only a few GPCRs, they have been selected by nature in higher organisms to play a pivotal role in signaling pathways. The GPCRs share highly conserved topologies with a perseverance of an analogous functional mechanism of action, though activation could be achieved by "ligands" as diverse as photons of light or the most complex polypeptide hormones3. They respond to, and transmit key physiological signals from the exterior of a cell to its interior, thereby influencing a host of biochemical processes. GPCR stimulation by ligands like hormones or neurotransmitters activates a GTP-protein coupled signal transduction pathway leading to a network of physiological responses. For the design of selective ligands for the various histamine receptor subtypes, it is important to know how histamine interacts with its receptor-binding site. Pharmacological and biophysical studies have provided a significant body of indirect evidence for the local environment of the bound histamine, without resolving the molecular detail of the agonist binding site, the conformation of histamine, or the molecular mechanisms of agonist activation4, 5. The currently proposed models for both the agonistic binding site on the histamine receptors and the accessory activation mechanism are controversial6, 7, 8. On the one hand models exist that are based on a tautomeric shift, which is thought to occur after histamine binds to the receptor: On one side of the imidiazole ring histamine accepts a proton, while on the opposite side it donates one. The suggested tautomeric shift of the imidazole moiety of histamine is supposed to trigger the receptor. However, several histamine receptor agonists have been developed which are not capable to undergo this tautomeric shift, but still behave as full agonists10. On the other hand there are models proposed11, 12, which indicate the importance of ion charges for the ligand recognition and receptor activation mechanism. In particular , modelling and mutagenesis studies have indicated that the imidazole ring can carry a full positive charge11, 12, because lorazepam 1 mg.
